Cell culture work is a mixture of logic considerations, traditional practices, and adaptations of general rules of thumb to the particular cell lines in use.

I strongly recommend to the novice to contact an extablished cell culture lab and to learn the first steps from an expert. If You wish (or if You have to) establish Your own local cell culture environment the tips given below may be helpful. Nonetheless they are no substitute for learning from the experts.

General principles and methods in conventional cell culture labs. Also equipment needed for permanent cell lines. This may not be used for the cultivation of fungi like yeast or bacteria. In addition to the things needed it may necessary to buy additional equipment according to local safety instructions.

Permanent cell lines are the most cultivated ones in conventional cell culture labs. The procedures and techniques can be learned in a few days if the newcomer is instructed appropriately. Also, the engagement required can be estimated. For example even on holidays You must be present....

Transient cell lines are a good alternative to stable cell lines if cell functions or structures are needed which are not accessible by conventional cells. In addition, the requirements to prevent contamination and the continuous work in holding a cell population alive are reduced.

Examples of different cell lines. The set is restricted to stable lines which can be purchased commercially. Only adherent cells are shown, because they express individual morphologies and are therefore interesting for microscopy.
